Experimental lens induced uveitis in rat

The present study was conducted to study the reaction in rat with their complete immu­nological check up at B Cell & `T' cell level. The results of study on rabbits have already been reported in the last conference 1978. The study on guinea pigs will be reported later.


  Material and method Top


The study was conducted on 40 outbred male wister rats. The twenty rats were given 6 mg of soluble hetero and homologous lens protein with complete Freunds adjuvant intramuscularly at every 10 days interval in 2 groups of rats. The serum from all animals was collected before and after immunisation and tested for antilens antibody. In total seven injections were given. The lenses were injured after last injection. Sera were collected and presence of antibodies were confirmed in all animals by agar gel diffusion, Haemagglutina­tion, Immunofluorescence & Immunoperoxi­dase techniques. During post needling period the eyes were regularly examined by Slit lamp microscope. The animals were sacrificed 6 days after needling. The blood was collected for se­rum antibody examination and for lymphocyte transformation test. The eyes were emmuclea­ted & histopathological study was done.

There were four control groups of 5 animals each in which the needling was done identi­cally.

a) Unsensitized animals.

b) animals receiving injection of complete Freunds adjuvant without any lens.

c) Animals receiving an injection of hetrologous lens without any adjuvant.

d) animals receiving injection of homolo­gous lens without any antigen.

All the animals received the injection at same time and at same regular interval. The needling was also done at the same time.


  Observations Top


Sera from all rats before immunisation were negative for lens antibodies as shown by simple agar gel diffusion, immunoelectropho­resis, passive haemagglutination, immunaflu­roscence & immunoperexidase tests. In all the rats where hom & hetrolegous lens were inje.c; ted with complete freunds adjuvant showed the presence of antilens antibody. The titre of anti­lens antibody varied from rat to rat but was alike as we observed in our study on rabbit, lymphoyte transformation index was variable and less changed in animals where heterologous lens was injected with complete Freunds adjuvant while in rats where homologous lens was injected with complete Freunds adjuvant showed the more rise in lymphoeyte transfor­mation index. The control animals neither showed any antilens antibody nor any effect on lymphocyte transformation index. The immunological part will be reported elsewhere.

Histology

In the eyes of rats where hetero & homo­logous lens was injected with complete Freunds adjuvant and antibody were also demonstrated showed prominent hyperaemia of iris, ciliary body with infiltration of anterior uvea by a small number of lymphocyte & plasma cells. The lenses were however infiltrated by poly­morph, cosinophils, lymphocytes, plasma cells and also presence of macrophages.

In few rats besides, there were also large histiocytes which contained debris of nuclear lens material. The reaction was much more suggestive of Arthus type of reaction.


  Discussion Top


Comparing to our study on rabbits on same work[1] the reaction in rats differed by being much more marked. The lens material was completely infiltrated by polymorphs, eosino­phils, lymphocytes, & plasma cells and occasional histiocytes. The reaction was more marked in rats where homologous lens when injected with complete Freunds adjuvant com­paring to the rats injected with heterologous lens along with same adjuvant. There was absence of multinucleated giant cells in any of rats. Marak[1] reported a granulomatous reac­tion in 40% of rats where homologous lens was injected while rats receiving heterologous lens with adjuvant showed only Arthus type of reaction. In our study we found severe reaction in all rats where homologous lens was injected comparing where heterologous lens was injected but'in both the group, the reaction was of Arthus type.


  Summary Top


The rats were immunised by hetero & homologous lens wilh complete Freunds adju­vant. The serum was tested for antilens antibodies & lymphocyte transformation index was found out by lymphocyte transformation test. The lens were needled & histopathologi­cal study of eye was done. In all rats severe Arthus type of reaction was observed.
